Twenty female C57BL/6 mice, 8–10 weeks of age, from Chongqing
Medical University Animal Center were used in this study. They were
housed under constant conditions at a temperature of 22 ± 2 °C, hu
midity of 50 ± 10 %, and 12 -h light/dark cycle. Female mice were
crossed with males in the evening and checked for vaginal plugs the
next morning. Additionally, their weights were recorded (embryonic
day 0, E0). On gestation day 14 (GD14), pregnant mice with weight
gain greater than 2 g were sacrifificed for primary mesenchymal cells. All
experiments abided by the Institutional Ethical Guidelines for
Experiments with Animals.
2.2. Cells and medium
Epithelial cells and medium were obtained from Shanghai Haling
Biotechnology Company, Limited. Primary epithelial cells were derived
from the palatal tissue of GD14 embryos from C57BL/6 mice.
Immunoflfluorescence was performed to detect the expression of the
epithelial marker ZO-1, and microscopic morphology was used to
confifirm its shape. A specifific medium was used to grow the epithelial
cells.
Mesenchymal cells were obtained from the palatal tissue of GD14
embryos from C57BL/6 mice. The methods of mesenchymal cell ac
quisition and culture were according to the previous study (Feng et al.,
2013). Immunoflfluorescence was performed to detect the expression of
the mesenchymal marker vimentin, and microscopic morphology was
used to confifirm its form. The media were DMEM/F12 (1:1) basic (1×)
(Gibco, USA) with 10 % fetal calf serum (FBS, Sijiqing, China).
